Three 24-hour recalls were administered by registered dietitians during telephone interviews. METASTATIC HEPATOBLASTOMA IN AN EQUINE NEONATE. A. T. Loynachan, D. C. Bolin, Livestock Disease Diagnostic Center, University of Kentucky, Lexington, KY. A 2-hour-old Thoroughbred neonate was submitted for necropsy with a clinical history of acute death following dystocia. Grossly, a 20 3 16 well demarcated multilobulated tan mass occupied and extended from the left liver lobe. Additional smaller masses were evident in the brain, choroid, meninges, and stylohyoid bone. Microscopically, the hepatic mass was separated from the normal parenchyma by a dense fibrovascular capsule, and was composed of sheets and cords of polygonal cells that were separated by sinusoid-like spaces, small clusters of fusiform cells, and loosely arranged mesenchymal regions. The polygonal cells stained immunohistochemically positive for alpha-fetoprotein, contained lipid and glycogen, and were consistent with fetal epithelial cells. The smaller fusiform cells were mixed throughout the polygonal cell population and were morphologically consistent with embryonal epithelial cells. The mesenchymal regions were composed of loosely arranged and well differentiated spindle cells. Intermixed throughout the mass were foci of necrosis and extramedullary hematopoiesis, and deposits of osteoid and bone. The brain, choroid, meninges, and bone masses were composed of similar fetal and embryonal epithelial cells. Based on the gross, microscopic, and immunohistochemical properties a diagnosis of primary mixed hepatoblastoma with teratoid features and multiple epithelial type metastases was made and chamomile.

In contrast, in incubated rat skeletal muscle, they appear to act by increasing the cytosolic concentration of citrate, an allosteric activator of ACC, as reflected by increases in the whole cell concentrations of citrate and malate [Saha, A. K., D. Vavvas, T. G. Kurowski, A. Apazidis, L. A. Witters, E. Shafrir, and N. B. Ruderman. Am. J. Physiol. 272 Endocrinol. Metab. 35 ; : E641E648, 1997]. We report here that sustained increases in plasma insulin and glucose may also increase the concentration of malonyl-CoA in rat skeletal muscle in vivo by this mechanism. Thus 70 and 125% increases in malonyl-CoA induced in skeletal muscle by infusions of glucose for 1 and 4 days, respectively, and a twofold increase in its concentration during a 90-min euglycemic-hyperinsulinemic clamp were all associated with significant increases in the sum of whole cell concentrations of citrate and or malate. Similar correlations were observed in muscle of the hyperinsulinemic fa fa rat, in denervated muscle, and in muscle of rats infused with insulin for 5 h. In muscle of 48-h-starved rats 3 and 24 h after refeeding, increases in malonyl-CoA were not accompanied by consistent increases in the concentrations of malate or citrate. However, they were associated with a decrease in the whole cell concentration of long-chain fatty acyl-CoA LCFACoA ; , an allosteric inhibitor of ACC. The results suggest that increases in the concentration of malonyl-CoA, caused in rat muscle in vivo by sustained increases in plasma insulin and glucose or denervation, may be due to increases in the cytosolic concentration of citrate. In contrast, during refeeding after starvation, the increase in malonyl-CoA in muscle is probably due to another mechanism. acetyl-coenzyme A carboxylase; malate; insulin resistance; hyperglycemia; hyperinsulinemia; glucose sensing.

As a result, industry has remained undaunted. For example, Coca-Cola purchased two companies that produced products containing ingredients that were the subject of warning letters to other companies.145 After taking over ownership of the companies, Coca-Cola continued to market most of the products as originally formulated.146 ONPLDS informed Coca-Cola of its concerns about these ingredients and suggested it follow the progress of an Agency review of popular herbals being used in foods. The GRAS review is being conducted by FDA's Office of Food Additive Safety.147 According to the project leader, there is no definitive date when the review will be done.148 Meanwhile, Coca-Cola continues to market products with the objectionable ingredients. In a revealing article published in a leading food industry trade publication, a senior FDA official admitted that the Agency would not be prosecuting companies whose products contained small amounts of ingredients not considered to be GRAS.149 This policy places the consumer in a "Catch 22" situation. It encourages companies to avoid prosecution for selling products with unapproved additives by selling products that contain misleadingly small amounts of the purported functional ingredient. Experimental procedure. Rats were anesthetized with an intraperitoneal injection of urethane 1.2 g kg in sterile water ; and their temperature was maintained at 37C using a homeothermic blanket. To perform intrathecal it ; catheterization, the rat head was fixed in a stereotaxic device and rotated nose downward. The catheter, a polyethylene tubing PE-10 ; stretched to 150% of its original length in hot water, was cut to the required length so that its distal opening reached either the L1-L2 or the L6-S1 segments of the spinal cord. For a female rat weighing 250 g, a 78-mm catheter was used to reach the L6-S1 segments. The skin and neck muscles were incised and retracted. The catheter was inserted into the subarachnoid space through a small hole made in the dura at the level of the atlanto-occipital membrane. It was pushed in the caudal direction and connected to a Hamilton syringe filled with NaCl 0.9%. The jugular vein was catheterized with a polyethylene tubing PE-10 ; filled with NaCl 0.9% and connected to a Hamilton syringe also filled with NaCl 0.9%. Rats were tracheotomized to facilitate spontaneous breathing. After a suprapubic midline incision, a small latex balloon, attached to a 1.5-mm-diameter catheter, was inserted through a small incision in the lumen of the left uterine horn and pushed caudally close to the cervix. Because of supplying difficulties in the course of our experiments, two different kinds of balloons were purchased from two different companies. The first kind Nicomed Amersham Medical Systems, 10.5 mm long ; , was used for all experiments of intrathecal lumbosacral and intravenous administrations of OT. The second kind Balt Extrusion, 9 mm long ; was used for all other experiments intrathecal thoracolumbar administration, [Thr4, Gly7]-OT, atosiban OT, HXM OT ; . It appeared that this second brand was markedly more sensitive than the first one, resulting in a greater frequency and amplitude of intrauterine pressure IUP ; rises measured. This was taken into account and statistical comparisons were made only with results obtained with the same type of balloon. The balloon and the catheter were filled with sterile water and connected to a pressure transducer Elcomatic 750, Phymep, France ; to monitor the IUP. Pressure of the balloon was increased and was maintained between 35 and 45 mmHg. Pressure signal was amplified Bionic Instruments, Phymep, France ; , sent to an analog-to-digital interface, sampled at a frequency of 5 Hz, and stored in a personal computer until offline analysis using Axotape software Axon Instruments, Union City, CA ; . An experiment consisted of the administration to the same rat of 10 l NaCl 0.9% followed by successive administrations of 0.3, 30, and 300 ng of either OT it or AVP it ; or [Thr4, Gly7]-OT it ; dissolved in 10 l NaCl 0.9%. Each administration including the initial one of 10 l saline ; lasted 10 20 s and was followed by a flush of 25 l NaCl 0.9%. Successive administrations were separated by a period of 15 min. In experiments with atosiban or HXM, a single administration of either compound was done before any other administration i.e., before the first administration of saline ; . All the rats of the present study underwent this protocol. Table 1 displays the different groups of rats used in the present study. At the end of the experiments, rats were killed with an overdose of urethane 3.6 g kg ip ; and perfused with saline through the left cardiac and charcoal.
Abstract Nuclear magnetic resonance spectroscopy was used to characterize the solution structure and backbone dynamics of a putative precursor form of -conotoxin MVIIA, a 25-amino-acid residue peptide antagonist of voltage-gated Ca2 + channels. The mature peptide is found in the venom of a fish-hunting marine snail Conus magus and contains an amidated carboxyl terminus that is generated by oxidative cleavage of a Gly residue. The form examined in this study is identical to the mature peptide except for the presence of the unmodified carboxy-terminal Gly. This form, referred to as -MVIIAGly, has previously been shown to refold and form its disulfides more efficiently than the mature form, suggesting that the presence of the terminal Gly may favor folding in vivo. The nuclear magnetic resonance NMR ; structure determination indicated that the fold of -MVIIAGly is very similar to that previously determined for the mature form, but revealed that the terminal Gly residue participates in a network of hydrogen bonds involving both backbone and side chain atoms, very likely accounting for the enhanced stability and folding efficiency. 15N relaxation experiments indicated that the backbone is well ordered on the nanosecond time scale but that residues 915 undergo a conformational exchange processes with a time constant of 35 microseconds.
